Strain Description ：
1.  The severe immunodeficiency of this strain is suitable for engraftment of human hematopoietic stem cells (HSC), peripheral blood mononuclear cells (PBMC), patient-derived xenografts (PDX) or adult stem cells … humanized experiments.
2.  The expression of theIl2rg (interleukin 2 receptor subunit gamma) gene is absent in this strain by flow cytometry. The presence of natural killer (NK) cells is extremely deficient (~1.8 %).
3.  Based on the background strain NOD.CB17-Prkdcscid/Narl, this strain is deficient in T cells and B cells.
4.  This CRISPR/Cas9-generated mutant of the Il2rg gene carries a deletion mutation from the start codon to the stop codon.

The Il2rg deletion mutant allele was created using CRISPR/Cas9 endonuclease-mediated genome editing. Guide RNAs were selected to target exon 1 and exon 8 of the Il2rg gene on chromosome X.These guide RNAs and Cas9 nuclease was introduced into zygotes of NOD.CB17-Prkdcscid/Narl, and then transferred to pseudopregnant females. Progeny were screened by genotyping of the target region. The founder was backcrossed to NOD.CB17-Prkdcscid/Narl for germline transmission.
